Technical Information
HiTrap™ HIC Selection Kit is ideal for small-scale HIC media screening and selection.
The kit includes HiTrap 1-ml columns containing the following media: Phenyl Sepharose™ High Performance, Butyl Sepharose High Performance, Phenyl Sepharose™ 6 Fast Flow (low sub), Phenyl Sepharose™ 6 Fast Flow (high sub), Butyl-S Sepharose™ 6 Fast Flow*, Butyl Sepharose™ 4 Fast Flow, and Octyl Sepharose™ 4 Fast Flow.
Phenyl Sepharose™ High Performance is based on a 34-μm matrix and is ideal for laboratory and intermediate process-scale separations and for final step purifications where high resolution is needed. The degree of substitution gives Phenyl Sepharose™ High Performance a selectivity similar to that of Phenyl Sepharose™ 6 Fast Flow (low sub).
Butyl Sepharose™ High Performance is based on a 34-?m matrix. The small beads give high resolution and are ideal for intermediate and final purification steps The relatively high ligand density enables high dynamic binding capacity for proteins, even at relatively low salt concentration.
Phenyl Sepharose™ 6 Fast Flow (low sub) and Phenyl Sepharose™ 6 Fast Flow (high sub) are based on a 90-μm matrix. They are ideal for initial and intermediate step purifications requiring a matrix with low to medium hydrophobicity. The availability of two degrees of substitution grades increases the possibility of finding the best selectivity and capacity for a given application.
Butyl-S Sepharose 6 Fast Flow is based on a 90-μm matrix. It is the least hydrophobic medium in the Fast Flow series. Butyl-S is suited for binding and elution of relatively strong hydrophobic molecules at comparatively low salt concentration.
Butyl Sepharose™ 4 Fast Flow is based on a 90-μm matrix. It is intended for initial and intermediate step purifications requiring a matrix with low to medium hydrophobicity. Butyl Sepharose™ 4 Fast Flow often works efficiently with relatively low salt concentrations. The mechanism of adsorption and desorption with the butyl ligand is different from that of the phenyl ligand, giving a different selectivity.
Octyl Sepharose™ 4 Fast Flow, which is also based on a 90-μm matrix, has a different hydrophobic character compared to the phenyl and butyl ligands, and is an important complement to the other hydrophobic matrices.
